In the identification of proteins,  dilute nitric acid is used to cover the food in the test-tube and then after heating the test-tube, water and ammonium hydroxide are added to it. If the food has protein, the colour would change into yellow first then on adding ammonium hydroxide, it will change from yellow to orange.
