The variegated leaf is kept in a dark room so that all the starch that was produced in the leaf gets used up. It is kept back again in the sunlight so that fresh starch is produced for the experiment and we get an accurate answer.

The plants are kept in dark to destarch them. Photosynthesis would not occur in absence of sunlight, so, plants are destarched by keeping in dark.
