Glycerine is put under a coverslip because it increases the refractive index of the sample.
Moreover in biological samples glycerine is "also used as" a dehydrating agent it prevents the moisture content of the sample and maintains it in a wet condition for dye to penetrate well.
In addition, it also reduces the air bubbles present in the sample so as to enable proper focussing of the sample in the microscope.
Glycerine can sometimes be used by drama artists to produce artificial tears.
