Chlorophyll is a green pigment in the leaf which must be removed for the iodine test..
If the chlorophyll is not removed the leaf is not decolourised and the leaf does not turn blue black when iodine is dropped .

chlorophyll is removed from the leaf before the test of starch because it needs to decolourised to observe the change to be observed
