Why facilated diffusion is sensitive to inhibitor that react with protein side chain

Facilitated diffusion involves passage through protein channels in the cell membrane - the side chains will be an essential part of"fixing" the channel protein in the hydrophobic core of the cell membrane. Reactions with these side groups will affect facilitated diffusion.
