Starch test is all about iodine test which is observed through change of color. Since, the chlorophyll is a green colored pigment in the leaves, so it becomes impossible to understand the color change due to iodine test for starch due to the presence of green pigments.

This is the reason; chlorophyll is extracted to de-colorize the leaves before testing the starch.

  • Greenish colour of chlorophyll can give a wrong indication of positive starch test.

Therefore, chlorophyll is removed by boiling in alcohol & then placing it in hot water before testing for starch.
