Humans have complex body designs.They have specialized cells for various activities.In unicellular organism - which are simple and their cells are in direct contact with environment or oxygen.
As humans have complex designs -- only the outer cells would acquire oxygen not the internal.So diffusion is insufficient to meet O2 requirements of humans.
