Why is distilled water used in preparation of culture media?

Distilled water is preferred in use, that is because the tap water may contain calcium and magnesium salts which can react with phosphate found in peptone or beef extract and produce calcium phosphate and magnesium phosphate salts which are water insoluble which can inhibit the growthof microorganisms.

- Tap water already contains various microbes that interfere with growth of desired microorganisms.
- It also contain impurities like calcium, magnesium, chlorine, fluorine which alter the composition of culture media.
- Ion content of tap water is variable and undetermined which makes it hard to define the characteristics of media.
- To prevent all this hazards of tap water, well purified distilled water is used to prepare culture media.
