IKI is normally light yellow in color; in the presence of starch it turns a deep blue. IKI can therefore be used to test for the presence and activity of the enzyme amylase, which breaks starch into maltose disaccharide units.

If we added saliva on starch, the salivary amylase present in saliva gradually acts on the starch and converts it into maltose.

Explanation:

  • Starch keeps giving blue colour with iodine till it is completely digested into maltose.
  • Addition of NaCl increases the rate of the reaction and catalyses the reaction better.
  • Thus, the hydrolysis of starch by salivary amylase increases.
  • It is the binding of one chloride ion at a specific site that induces the allosteric activation of the salivary amylase
