• Take a fresh leaf that must be green and alive and carefully with a blade, cut off a really small outer layer of the leaf's structure.
• Take a glass slide and put that peel on it. Add the stain as you like and glycerine too.
• Now cover it with a watch glass.
• Now take a microscope and adjust the mirror to allow sufficient light to enter the microscope, by watching through the eye piece.
• Now, as you like, rotate the various screws until you are able to see the required observation.

1.cut a small piece from the leaf.
2.peel off the cuticle from ventral surface of leaf using forceps and needle.
3.keep the leaf in petridish.
4.put 2-3 droops of safranin stain. Remove excess of stain using water.
5.apply glycerin on glass slide.
6.transfer the leaf to glass slide using needle and foreceps
7.put cover slip gently to avoid air bubbles formation .
8.observe the slide under light microscope.
