Functional Gene Cloning

Functional cloning is an approach for gene identification that exploits what is known of the protein whose dysfunction is associated with disease without knowledge of the chromosomal location of the gene . The amino acid sequence of the protein (or peptides derived from the protein of interest) is used to predict the gene coding sequence, and cDNA libraries are screened with an oligonucleotide probe (degenerate oligonucleotides). In addition, polymerase chain reaction can be employed to amplify the cDNA using oligonucleotides designed from the amino acid sequence . Functional cloning has been used to identify genes causing human diseases such as phenylketonuria and sickle cell anemia
