Because,

You never use "Despite of". You should use "Despite her illness",as given in the sentence.

"Of" is most commonly used while using "Inspite".

For example,

Inspite of her injury,she went to the school.

Notice that "of" is followed after Inspite,and NOT after DESPITE.
