Acetyl-CoA, main product of the Pyruvate Dehydrogenase Complex in aerobic respiration, starts the Krebs cycle. (An irreversible reaction that islink between glycolysis and the citric acid cycle.) The mechanism of the synthesis of acetyl coenzyme A from pyruvate requires five coenzymes and three enzymes.

- Pyruvic acid formed in glycolysis is taken as input and acetyl Co-A is formed which is further used as substrate in Krebs cycle.
- This reaction occurs in mitochondria.
- Here, pyruvate undergoes oxidative decarboxylation to form acetyl group.
Pyruvate --> acetyl group + CO2
- This acetyl group combines with coenzyme A to form acetyl co-A.
Acetyl group + Co-A --> Acetyl Co-A
- NAD combines with hydrogen to form NADH2.
NAD +2 H+ --> NADH2
- Enzyme employed here is pyruvate dehydrogenase complex.
- Overall reaction is -
Pyruvate + Co-A + NAD --> Acetyl Co-A + CO2 + NADH2
