Gram-positive bacteria do not have an outer cell membrane found in Gram-negative bacteria. The cell wall ofGram-positive bacteria is high in peptidoglycan which is responsible for retaining the crystal violet dye.

It is difficult to treat gram-negative bacteria in comparison to gram-positive bacteria due to following reasons. There is a membrane present around the cell wall of gram-negative bacteria which increases the risk of toxicity to the host but this membrane is absent in gram-positive bacteria.

In some bacteria, cell membrane consists of two layers while in some there are three. ... The difference between Gram-positive and Gram-negative bacteria is the thickness of the peptidoglycan cell wall. Gram-positive bacteria has a thick layer of peptidoglycan, while Gram-negative has a thin layer.
