DNA Polymerase copies in the direction of 5' to 3' but the lagging strand runs in the direction of 3' to 5' so the replication in the lagging strand happens in fragments laid by RNA Primers(called Okazaki fragments).These fragments are joined together by DNA Ligase. Hence the replication of this strand is discontinuous.

Whereas the leading strand runs in the same direction as the direction required for replication by DNA Polymerase(5' to 3') so the replication is continuous.

  • At the beginning of DNA replication, both strands f the DNA is separated by enzyme helicase.
  • A new replication fork that can move in the direction of unwinding DNA is formed.
  • Short RNA primer is produced near the replication fork.
  • It is then extended by DNA polymerase.
  • DNA polymerase can replicate DNA in 5'-3' direction.
  • 3'-5' is called the leading strand.
  • But the template strand with 5'-3' polarity is synthesized discontinuously because it is in the opposite direction of polymerization of the replication fork.
  • The inherent opposite polarity of the two strands is the reason for the continuous and discontinuous DNA synthesis.
