Gram-staining

Explanation:

  • Gram-staining is a technique which is used to differentiate between two groups of bacteria on the basis of staining of cell wall components.
  • The gram-staining procedure distinguishes between gram positive and gram negative bacteria depending upon the coloring of the cell walls as red or violet.
  • The gram-staining technique was invented by Hans Christian Gram.
  • The procedure is as follows:
  1. Apply the primary stain (crystal violet) and heat fix the smear.
  2. Add iodide, which binds to the crystal violet and traps it in the cells.
  3. Decolorize the stain by using ethanol.
  4. Apply counterstain with safranin.
  • The gram positive bacteria will take up the crystal violet stain will be colored as violet and the gram negative bacteria will take up the safranin stain and will be colored as red or pink.
