Pleuromutilin (Tiamulin)

The only commercial antibiotic produced by a basidiomycete is the diterpene pleuromutilin. Pleuromutilin was first isolated from Pleurotus mutilus and Pleurotus passeckerianus in a screening for antibacterial compounds. Pleuromutilin is active against Gram-positive bacteria, but its most interesting biological activity is its effectiveness against various forms of mycoplasms. The preparation of more than 66 derivatives of pleuromutilin resulted in the development of tiamulin, which exceeds the activity of the parent compound against Gram-positive bacteria and mycoplasms by a factor of 10–50. The minimal inhibitory concentrations against different strains of mycoplasma were in the range 0.0039–6.25 μg ml−1.
