Your answer is thionin.

      Thionin is used to observe amoeba or paramecium under microscope, in a water drop on the glass slide.

      As thionin is a LTP (Lipid transfer protein) they can do act directly any movement to be recognised. So that, they can used as stain to see amoeba in presence of water drop on glass slide. Similarly, methelyne blue belongs to this property.

    In fact, most known function of methelyne blue is to be used as stain to observe animal cells like amoeba and paramecium more clearly through hybridization membranes like water and make their nucleus visible out clearly.
