What is staining?why some regions appear darker than other regions after staining?

staining is basicially a dye which use to color the blood component .nucleous appear darker because of the acidic nature of the nucleaous which commonly stained by basic dye .example ..methylene blue dye
and acidic dye(eosin dye) which give color to basic component i.e. cytoplasm
