Which of the following two graphs will be a straight line at constant temperature?: P versus V or P versus \frac{1}{V} (at constant T) P versus \frac{1}{V} graph will be a straight line.

According to the Boyle’s Law,

                                                   PV = K (a constant)

where P = Pressure, V= Volume.

which implies P is inversely proportional to the Volume.

Thus this graph P (Pressure) versus 1/V (V=volume) is a straight line at the constant T (temperature).

whereas the graph P (Pressure) versus V (V=volume) is not a straight line at the constant T (temperature), since P will be directly proportional to the Volume.
